HuGENet's HuGE Review Handbook provides guidance for undertaking HuGE reviews. The intention of the Handbook is to help investigators to make sensible decisions about the methods they use, rather than to dictate arbitrary standards. It also offers a framework to ensure that HuGE reviews have a similar, easy-to-follow, format. The Handbook is an evolving, online document that is updated over time as methodology and understanding develops.

The Handbook resulted from a methodology workshop held in Cambridge, UK, in November 2004. The workshop brought together epidemiologists, geneticists, statisticians and other healthcare researchers to develop methodological guidance for authors of systematic reviews and meta-analyses in human genome epidemiology, and to identify any potential developments that could improve their validity. The Handbook is edited by Julian Little and Julian Higgins, on behalf of a writing group consisting of Molly Bray, Julian Higgins, John Ioannidis, Muin Khoury, Julian Little, Teri Manolio, Liam Smeeth and Jonathan Sterne.

The Handbook is modelled in part on the Cochrane Handbook for Systematic Reviews of Interventions, which provides even more detailed advice for people undertaking systematic reviews.
